The mechanism of drug release from tablets with insoluble membranes is that digestive juices can quickly penetrate the pores of the membrane and dissolve the active substance of the tablet or cause it to swell. In the first case, the dissolved substance diffuses through the film in the opposite direction - towards the gastrointestinal tract under the influence of the difference in concentrations, in the second - the membrane breaks due to the increase in tablet volume, after which the drug is released in the usual way. The uniform release of the drug substance through the intact membrane occurs regardless of the composition, enzyme action and pH of the medium in the gastrointestinal tract. It depends only on the solubility and pore size of the shell, which can be modified as necessary. Therefore, it is possible to obtain tablets with a pre-calculated rate of drug release.

Water-soluble coatings improve the appearance of tablets, correct their taste and smell, protect against mechanical damage, but do not protect against moisture from the air. Water-soluble membranes form PVP, MC, Na CMC, etc., applied to tablets in the form of water-ethanol or aqueous solutions. Coatings soluble in gastric juice - films that protect against moisture, provide rapid dissolution in the stomach (within 10-30 minutes). The film-forming agent of this group includes polymers having diethylaminomethyl cellulose, sugar para-aminotenzoates, and cellulose acetate in the amino group molecule. The tablets are coated with solutions of these substances in organic solvents - ethanol or isopropanol, which volatilize during coating and drying. Enteric coatings protect the drug substance contained in the tablet from the acid reaction of the gastric juice, protect the gastric mucosa from the irritating effect of certain drugs, localize the drug substance in the intestine, prolonging its action to a certain extent, and also have a pronounced moisture protective effect. The process of dissolution of the membranes in the body is due to the action of a complex of enzymes on them to various solubilizing substances contained in the intestinal juice.

As film-forming agents for the preparation of enteric coatings, high molecular weight compounds are used. They dissociate in a neutral and alkaline environment with the formation of insoluble salts. Both natural substances are used: shellac, casein, keratin, paraffin, ceresin, cetyl alcohol, and synthetic products: stearic acid in combination with fats and bile acids, butyl stearate, acetylphthalyl cellulose, polyvinyl acetate flatate, polyacrylic resins (Eudragid). The listed film-forming agents are applied to tablets in the form of solutions. To obtain colored membranes, pigments and dyes are added to the solutions. By varying various polymers and different film thicknesses, it is possible to achieve tablet disintegration in a specific section of the intestinal tract. Enteric coatings withstand the effects of gastric juice for 2-4 hours or more, which allows such dosage forms to pass through the stomach unchanged; in the intestinal juice, they disintegrate within 1 h, providing the release of the drug substance in the intestine. Insoluble coatings are microporous films for the controlled release of a drug substance. The main purpose of this type of coating is to protect tablets, pellets or granules from mechanical damage and from exposure to atmospheric moisture, eliminate the unpleasant odor and taste of the drug substance, and prolong its action.
