Preparation of suppository base

The stage of preparation of the suppository base begins with the sizing of its components. In the first stainless steel reactor with a steam jacket melt paraffin, in the second reactor melt hydrofat by supplying steam into the steam jacket. The heated hydrofat is transferred to the reactor with pre-melted paraffin by means of a pump. The mixture is heated to a temperature of 60-70 ° C and cocoa butter is added. In order to avoid changes in the modification of cocoa butter, heating should not exceed 70 ° C and be prolonged. After complete melting of the base it is stirred for 40 min with a stirrer. The melting point and the time of complete deformation are determined in the finished base.

The preparation of the suppository mass is carried out in a reactor, where the ready base is fed with compressed air and drug substances are added. As an example, the LT series reactor-mixer manufactured by the Italian company ZUMA s.r.l. is used. Mixers of this series are designed for the preparation of suppository mass, creams, ointments, etc. and are available in four sizes with the volume of the working tank 30, 60, 140 and 250 liters. The working tank of the reactor is a polished stainless steel tank with a water jacket for heating and melting of the mixture. The tank can be rotated by 90° for easy draining and washing. The agitator blades of the reactor are equipped with scraper blades. All working parts of the apparatus are made of stainless steel. The mixer reactor is equipped with safety guards and is manufactured according to international standard.

Technical specifications

If the melting temperature of the base is higher or lower than the specified one, then paraffin or hydrofat is introduced into the base heated to 60-70 ° C with thorough mixing. The base is filtered through a druk-filter using a brass mesh or belting, analyzed by melting temperature, solidification and time of full deformation and transferred to the apparatus department.
Druk filters are pressurized capacitive filters. Druk-filters are vertical vessels (usually with a jacket) with an elliptical bottom.
The vessel filter housing can be open or closed. The upper part of the housing is fed with the slurry to be separated. The filtrate is discharged from the lower part. During filtration, compressed gas is fed into the slurry-filled housing.

Additional information

The filter baffle is located on the false bottom. The resulting sludge is washed, dried and discharged by means of an agitator through a hatch located on the side of the housing or in the center of the false bottom. In some designs, the sludge is removed after lowering the false bottom by moving the filter cloth belt, which passes through the regeneration chamber at the end of the sludge discharge. Druk filters are known, the body of which can be rotated by 90-180° in the trunnions fixed on its cylindrical part. Druk filters are used to separate medium and well-filtered suspensions in medium and low-tonnage productions.
