Dry granulation is a method in which powdered material (mixture of drug and excipients) is subjected to compaction to obtain granulate. Dry granulation is used when wet granulation affects the stability and/or physicochemical characteristics of the drug substance, and when the drug and excipients do not compress well after the wet granulation process.
If drug substances undergo physical changes (melting, softening, color change) or chemical reactions during drying, they are briquetted, i.e. briquettes are pressed from the powder on special briquetting presses with large-sized dies (25×25 mm) under high pressure.

In the second step, the agglomeration process takes place due to the formation of solid bridges as a result of sintering of particles, partial melting or crystallization of soluble substances. Next, solid bridges are formed between the particles through chemical reaction, solidification of binders or crystallization of insoluble substances. The dry granulation process is carried out on special equipment. The combined plant combines the processes of compacting, crushing and separating the resulting granules. The principle of operation of the press-granulator is as follows: rotating in different directions, the rollers grasp the powdery mixture and press it through the holes in the wall of the hollow rollers. Inside the hollow rollers, a knife cuts the resulting pellets.

The resulting briquettes are crushed using mills, fractionated using sieves and pressed on tablet machines into tablets of a given weight and diameter. Nowadays, using the method of dry granulation, dry binders (e.g. microcrystalline cellulose, polyethylene oxide) are introduced into the composition of the tablet mass, providing under pressure the adhesion of both hydrophilic and hydrophobic particles. The bonding of particles to each other occurs under the influence of forces of different nature. At the first stage molecular, electrostatic, magnetic forces act. Then the formation of bonds between particles takes place, after which capillary forces begin to act.
