Binders for wet granulation

The most common and effective wet granulation binder in the modern pharmaceutical industry is a synthetic polymer such as Collidon (PVP), the various grades of which (Collidon 25, 30 and 90F) are widely available on the market.

Plasdon polymers have excellent bonding ability, good film forming properties, surfactant properties and high solubility in water and many solvents used in pharmaceutical applications. Due to this combination of properties, these polymers are widely used in a number of pharmaceuticals. Plasdone polymers have long been used as binding agents in wet granulation. Plasdone®S-630 Copovidone is a synthetic 60:40 linear polymer of N-vinyl-2 pyrrolidone and vinyl acetate. With its unique properties, Plasdone S-630 is well suited as a tablet binder for direct pressing and dry granulation applications, as well as a wet granulation binder.

Technical specifications

The granules obtained with PVP are hard, easy to bulk, forming harder tablets with low brittleness. PVP polymer improves the solubility of the active ingredient by forming complexes. In addition, PVP acts as a crystallization inhibitor. The regularities presented below were identified by BASF’s fine chemistry department. They are discussed using Collidon as an example. Wet granulation using Collidon 25, Collidon 30 and Collidon 90F generally results in a harder granular material with better bulk properties than other binders – less abrasion and higher bond strength. The polymer content of the granulating fluid used can vary from Collidon 25 – 2-5%; Collidon 30 – 2-5%; Collidon 90F – 1-3%.

Thus, BASF’s research showed that Collidon 90F produced the highest tablet hardness at the same pressing pressure. All binders were used in the amount of 3%. Based on the data presented, it can be concluded that the increased hardness and reduced brittleness of calcium phosphate tablets when Collidon 30 is used as a binder in the wet granulation process. In addition to Collidon, there are a large number of substances used as binders in the pharmaceutical industry. Let us consider two of them.
